Genetic analysis of the first mcr-1 positive Escherichia coli isolate collected from an outpatient in Chile BJID
Gutiérrez,Camila; Zenis,Javier; Legarraga,Paulette; Cabrera-Pardo,Jaime R.; García,Patricia; Bello-Toledo,Helia; Opazo-Capurro,Andrés; González-Rocha,Gerardo.
ABSTRACT Global dissemination of mcr-like genes represents a serious threat to public health since it jeopardizes the effectiveness of colistin, an antibiotic used as a last-resort treatment against highly antibiotic-resistant bacteria. In 2017, a mcr-1-positive isolate of Escherichia coli was found in Chile for the first time. Herein we report the genetic features of this strain (UCO-457) by whole-genome sequencing (WGS) and conjugation experiments. The UCO-457 strain belonged to ST4204 and carried a 285 kb IncI2-type plasmid containing the mcr-1 gene. Moreover, this plasmid was transferred by conjugation to an E. coli J53 strain at high frequency. The isolate harbored the cma, iroN, and iss virulence genes and did carry resistance genes to...

Genetic heterogeneity of Escherichia coli strains isolated from raw milk, Minas Frescal cheese, and food handlers Arq. Bras. Med. Vet. Zootec.
Campos,M.R.H.; André,M.C.D.P.B.; Borges,L.J.; Kipnis,A.; Pimenta,F.C.; Serafini,A.B..
From February 2004 to March 2005, 140 samples of food handlers - hands and nostrils - (92), raw milk (24), and minas frescal cheese (24) were analyzed for the presence of Escherichia coli in a dairy processing plant of Goiás State. Forty-seven E. coli strains were obtained and compared by DNA macrorestriction patterns obtained from pulsed-field gel electrophoresis following XbaI restriction in order to investigate the possible sources of cheese contaminations. Based on PFGE genotyping, one strain isolated from food the hands of a handler and five strains isolated from raw milk were identical or closely related to six strains from cheese suggesting, in these cases, the probable source of E. coli contamination in cheeses. No strain isolated from the nostrils...

Glucuronidase activity of Escherichia coli isolated from chicken carcasses BJM
Perin,Luana Martins; Yamazi,Anderson Keizo; Moraes,Paula Mendonça; Cossi,Marcus Vinícius Coutinho; Pinto,Paulo Sérgio de Arruda; Nero,Luís Augusto.
To identify Escherichia coli through the production of β-D-glucuronidase (GUD), 622 suspect cultures were isolated from chicken carcasses and plated in PetrifilmTM EC. Of these cultures, only 44 (7.1%) failed to produce GUD. This result indicates the usefulness of GUD production for estimating E. coli populations in chicken.

Heterologous expression of an insecticidal gene from the armed spider (Phoneutria nigriventer) J. Venom. Anim. Toxins incl. Trop. Dis.
Figueiredo,J. E. F.; Kalapothakis,E.; Gomez,M. V.; Bressan,W..
Insect-pests are global problems that cause severe damage to crop plants, and their control is commonly based on chemical insecticides. However, negative effects of pesticides on the environment and human health emphasize the necessity to develop alternative methods for insect-pest control. In the present study, a gene coding for the insecticidal peptide TX4(6-1) of the Brazilian armed spider (Phoneutria nigriventer) was cloned in fusion with maltose binding protein (MBP) and expressed in Escherichia coli. The affinity purified protein MBP-GlyTX4 was cleaved with the Xa factor and used for a bioassay against Spodoptera frugiperda and rabbit immunization. Five micrograms GlyTX4 protein injected into the hemocoel of larvae and abdominal cavity of adults...

Human sepsis-associated Escherichia coli (SEPEC) is able to adhere to and invade kidney epithelial cells in culture BJMBR
Conceição,R.A.; Ludovico,M.S.; Andrade,C.G.T.J.; Yano,T..
The adhesins of extraintestinal pathogenic Escherichia coli are essential for mediating direct interactions between the microbes and the host cell surfaces that they infect. Using fluorescence microscopy and gentamycin protection assays, we observed that 49 sepsis-associated E. coli (SEPEC) strains isolated from human adults adhered to and invaded Vero cells in the presence of D-mannose (100%). In addition, bacteria concentrations of approximately 2 x 10(7) CFU/mL were recovered from Vero cells following an invasion assay. Furthermore, PCR analysis of adhesin genes showed that 98.0% of these SEPEC strains tested positive for fimH, 69.4% for flu, 53.1% for csgA, 38.8% for mat, and 32.7% for iha. Analysis of the invasin genes showed that 16.3% of the SEPEC...

Identification of putative new Escherichia coli flagellar antigens from human origin using serology, PCR-RFlP and DNA sequencing methods BJID
Tiba,Monique Ribeiro; Moura,Claúdia de; Carazzolle,Marcelo Falsarella; Leite,Domingos da Silva.
Escherichia coli has been isolated frequently, showing flagellar antigens that are not recognized by any of the 53 antisera, provided by the most important reference center of E. coli, The International Escherichia and Klebsiella Center (WHO) of the Statens Serum Institute, Copenhagen, Denmark. The objective of this study was to characterize flagellar antigens of E. coli that express non-typeable H antigens. The methods used were serology, PCR-RFLP and DNA sequencing. This characterization was performed by gene amplification of the fliC (flagellin protein) by polymerase chain reaction in all 53 standards E.coli strains for the H antigens and 20 E. coli strains for which the H antigen was untypeable. The amplicons were digested by restriction enzymes, and...

Improvement of penicillin G acylase expression in Escherichia coli through UV induced mutations BJM
Arshad,Rubina; Farooq,Shafqat; Ali,Syed Shahid.
We used ultraviolet (UV) radiation to induce mutation in three locally isolated strains of Escherichia coli. Different dilutions of bacterial cultures were exposed to UV lamp of 254 nm wavelength for different time intervals at varied distances ranging from 5 to 210 sec and 5 to 100 cm. Viable colonies were screened for mutants with an increased production of penicillin G acylase (PGA) and a reduced production of β-lactamase, which are the desired properties of PGA producing industrial strains. A survival curve was made to get optimum exposure time and distance. The survival percentage for each exposure period was calculated and 1-5% survival was found useful for obtaining mutants with desired change. Screening for PGA and β-lactamase constitutive and/or...

In vitro inhibition of adhesion of Escherichia coli strains by Xylitol BABT
Silva,Annelisa Farah da; Suzuki,Érika Yoko; Ferreira,Aline Siqueira; Oliveira,Murilo Gomes; Silva,Sílvio Silvério da; Raposo,Nádia Rezende Barbosa.
The present study aimed to evaluate xylitol's antimicrobial and anti-adherence activities on Escherichia coli (ATCC 8739) and on another clinical strain enteropathogenic E. coli (EPEC). In vitro minimum inhibitory concentration (MIC) test and adhesion assays were performed using 0.5, 2.5 and 5.0% xylitol. It was found that xylitol did not have antimicrobial properties on these strains. The scanning electron microscopy (SEM) demonstrated that the slides treated with xylitol had a significant reduction in the number of bacilli and the inhibition of microbial adhesion was probably the xylitol's mechanism of action. Xylitol could be a possible alternative on the control of E. coli infections.

Costa,João Carlos Miguel; Espeschit,Isis de Freitas; Pieri,Fábio Alessandro; Benjamin,Laércio Anjos; Moreira,Maria Aparecida Scatamburlo.
Bacterial biofilms are involved in the aggravation and recurrence of clinical mastitis in dairy herds. Several factors such as pH, temperature, concentration of O2 and glucose can affect their induction and growth rates. In this study, biofilm production was demonstrated by 27 Escherichia coli strains isolated from bovine mastitis at different pH values depending on the availability of glucose, mimicking conditions found in mammary glands affected by the disease. Biofilm formation was analyzed by spectrophotometric analysis in microtiter plate with 16 different culture media and by scanning electron microscopy. Biofilm formation was greater in isolates cultured under conditions associated with low glucose availability (0.5% or 1.5%) and with either an...

Vaz,Michelle Rossana Ferreira; França,Ricardo Luiz Soares de; Andrade,Sirtys Santos Lessa de; Sousa Junior,Francisco Canindé de; Santos,Everaldo Silvino dos; Martins,Daniella Regina Arantes; Macedo,Gorete Ribeiro de.
With the advent of recombinant DNA technology, recombinant protein expression has become an important tool in the study of the structure, function and identification of new proteins, especially those with therapeutic functions. Escherichia coli has been the predominant prokaryote used in genetic engineering studies due to the abundance of information about its metabolism. Despite significant advances in molecular biology and immunology of infections, there are as yet no prophylactic drugs capable of preventing visceral leishmaniasis. It is therefore important to identify specific antigens in order to develop vaccines and diagnostic kits against this disease. The objective of this study was to evaluate the influence of culture medium on the production of...

Investigating an avian source of shellfish faecal contamination in the Thau lagoon (the Mediterranean, France) ArchiMer
Derolez, Valerie; Serais, Ophelie; Caprais, Marie-paule; Le Saux, Jean-claude; Messiaen, Gregory.
Microbes regularly contaminate Thau lagoon shellfish. Between 1997 and 2007, 18% of the analyses carried out in dry weather recorded more than 230 Escherichia coli / 100 g of bivalves (Ifremer French microbial monitoring network), leading the administration to restrict the shellfish harvest area, including suspension of production. This study aims to determine, the contribution of seabirds, which roost at night on the Thau lagoon shellfish growing frames, to shellfish faecal pollution in dry weather.Data were collected in 2008 from both the drainage area (pollution input) and the lagoon (water quality and microbial monitoring, seabird counts) in dry weather. The data showed that, with no watershed input or lagoon resurgence, microbial contaminants were...

Is nosocomial Escherichia coli bacteremia a predictive risk factor for mortality? BJID
Tuon,Felipe F.; Rocha,Jaime L.; Guadagnin,Fernanda A.; Alves,Márcia S..
The objective of this study was to determine risk factors associated with mortality in patients with nosocomial Escherichia coli bacteremia from January 2009 to January 2011. In a retrospective study the medical records of 88 patients over 18 years with nosocomial bacteremia caused by E. coli were analyzed. In univariate analysis several risk factors, including chronic renal failure, altered mental status, leukocytosis, and higher Charlson index of comorbidities were associated with mortality. In multivariate analysis only altered mental status remained independently associated with mortality. Mental confusion can be a risk factor for mortality in patients with E. coli bacteremia.
